Test
Testing the lens requires specific lens test charts, preferably on a white background, and a tripod. You need to know the measurements of your test board area, as well as the magnification value used when photographing the charts.
Results
Viewing the photos at 100 percent with image editor software lets you analyze the performance of the lens, to see how well it detected the faintest lines on the chart. The black and white lines -- which should be distinguishable from one another -- determine the horizontal and vertical directions. The numbers indicate the lines per millimeter that the lens detected.
Calculations
Multiplying the horizontal lines per millimeter value by the magnification used reveals your resolution for the lens. The calculation is repeated for vertical.

For optimal results, you need to test the lens at different apertures and distances. The more tests conducted, the more accuracy and understanding of the lens gained. Image resolution isn't based on one factor, but rather a combination of the lens and the camera sensor. Testing for both allows for better resolution.
